EGFR Effector Reporter Cell
I. Background | |
EGFR(Epidermal Growth Factor Receptor)是上皮生長(zhǎng)因子(EGF)細(xì)胞增殖和信號(hào) 傳導(dǎo)的受體,EGFR 屬于 HER 受體家族,該家族由四種相關(guān)蛋白 EGFR(HER1/ErbB1)、 ERBB2(HER2)、ERBB3(HER3)和 ERBB4(HER4)組成,屬于激酶型受體, 位于細(xì)胞膜表面,靠與配體結(jié)合來激活。EGFR 與腫瘤細(xì)胞的增殖、血管生成、腫瘤侵襲、 轉(zhuǎn)移及細(xì)胞凋亡的抑制有關(guān)。 HER 受體通過與不同的配體結(jié)合而被激活,包括 EGF、TGFA、肝素結(jié)合 EGF 樣生長(zhǎng) 因子、雙調(diào)蛋白、β細(xì)胞蛋白和上皮調(diào)節(jié)蛋白。配體與受體的細(xì)胞外結(jié)構(gòu)域結(jié)合后,受體形 成功能活性二聚體[EGFR-EGFR(同型二聚體)或 EGFR-HER2、EGFR-HER3、EGFR- HER4(異二聚體)]。二聚化誘導(dǎo)激酶結(jié)構(gòu)域的激活,從而導(dǎo)致多個(gè)殘基上 的受體自磷酸化。從而導(dǎo)致一系列接頭蛋白(如 SHC、GRB2)的募集,并激活一系列細(xì) 胞內(nèi)信號(hào)級(jí)聯(lián)反應(yīng)以影響基因轉(zhuǎn)錄,進(jìn)而導(dǎo)致癌細(xì)胞增殖、減少細(xì)胞凋亡、侵襲和轉(zhuǎn)移, 并刺激腫瘤誘導(dǎo)的血管生成。 | |
